I'm Venus in this aspect. It's a male friend whose Moon squares my Venus exact. It feels very sweet, when we're together, we can't stop laughing. He feels really close to me, like we can always trust each other.

But to tell the truth, i think that this situation is not what usually this aspect would give as we also have his Venus exactly conjunct my Moon, his Moon also conjuncts my Neptune and his Mercury conjuncts my Sun... Not to mention his nodal axis exactly on my ASC/DSC + his ASC exactly conjuncts my IC, his Jupiter conjuncts my MC exact and my Mercury and Venus conjunct his MC exact... Big time! No way to separate this single aspect Venus square Moon exact...

What if there is Moon conjunct Venus the other way around? Who feels unappreciated - Moon or Venus in a square aspect?

Moon conjunct Venus would be the reverse - appreciation of each other on a basic level.
It's supposedly one of the sweetest aspects in synastry (makes sense, who doesn't like being appreciated?).
In your case, I'm sure that compensates.

I think the Moon person would feel unappreciated by the Venus person more than vice versa.

ETA: although it probably goes both ways.

This. My husband is the Moon, and I am the Venus. The square is exact.

We show our appreciation in completely different ways, and sometimes it leads to misunderstandings or feeling unappreciated by the other person (both ways, although I think that my husband feels it more than I do, as my Venus is quincunx my Moon).

But we're alright. We love each other very deeply. Other aspects count, too.

My Venus is trine his Vertex/Venus, and his Sun/Venus are conjunct my AC. Moon trine Moon. Not so bad.

You may appreciate this example, in that I have this aspect with someone who would not be a priority to me by herself, but is the girlfriend of a friend.

FWIW I am the Venus (Aries) person and she the Moon (in Cancer).

We've already had an incident where either (probably) she misunderstood me and reacted quite emotionally although somewhat cryptically as to the source of her anger; or, her outburst wasn't about me at all, LOL (either way, one of us misinterpreted something). You can really see how the signs involved affect the particulars of the dynamic - scenario A) she thought I was being aggressive (Aries) and was sensitive to it (Cancer), or B) she was indirect(Cancer) about the source of her emotions, and I just assumed it was about me (Aries).

It semi-resolved itself; her demeanor lightened up, she responded positively to the thing that initially seemed to set her off. I suspect my friend intervened and changed her perspective. (I say semi-resolved because obviously I haven't initiated any communication about all this, because of additional factors besides this aspect. If my friend were my child or other much closer person instead, I would likely have been more proactive about getting clarity on what happened.)

Shortly after that incident, though, me and Moon-person had a brief supportive email exchange (which I initiated) as my friend is going through a rough time. It was actually very warm, but very polite, suggesting we are careful with each other.

We have only a friendship on a work level. In other words, we talk, it's personal, but we don't hang out or call each other type of stuff.
Maaaaaaaan is it sweet though. I've never felt so content and appreciative around someone. Just loving their company.

Now the square, nothing comes to mind as far as experiencing it myself.

The way I could see it play out is the moon person exhausting themselves on the venus person, eventually feeling an imbalance in the relationship to the venus person. Becoming upset that they don't seem to be getting the same in return.

Maybe I've even got that backwards but in general I think that's the type of thing that would happen. It might start out all smiles and end up all misunderstood.

It is only one aspect though. There are others that could easily offset it.

This is absolutely how it plays out between my husband and I. I have to consciously work to try to balance the give/take in our relationship, because it can easily become unbalanced (in my favor). I think that if the Venus person is willing to try to balance things out by giving more of themselves the energy can be worked with. But yeah, other helpful aspects from Venus and/or Moon are always a good thing.
