I don't think anything'll flat-out doom a synastry.

For every example that people would give, there would be an example of a couple which has done absolutely fine with it.

If you refer to "star-crossed lovers", I've read that -for instance- contacts in the SN of one or another (or both) may indicate such "doomed" relationship.

In general, there aren't any hard and fast rules that can apply in all cases. Synastry and composites charts are wholes not to be judged by one or two aspects. There's free will too.
