So the chart ruler is it the ruling planet of the ascendant? What kind of influence does it have in synastry?
What about if a man has his sun conjunct the ruler of his ascendant? Will his sun 'act' more like the ascendant?

Yes, the modern way of finding a chart ruler is going by the Asc ruler.
