My favorite comedic line of recent times is:

"Rehtard"

It's funny because it's ironic. Not only is the character saying it of questionable intelligence, but he also manages to screw up the pronunciation. It's a perfect one-worder much less one-liner.

Do you know:
"... we're friends with every creature coming down the pipe, in fact we've never met an animal we didn't like ..."
OK it's a song so ...
But anyway, the context makes it a classic!

And this one which I use every now and then..

"Don't eat the green ones, their not ripe yet"

"And they have no purpose that unites them, so they just drift around, blundering through life until they die. Which they-they know is coming, yet every single one of them is surprised when it happens to them. They're incapable of thinking about what they want beyond the moment. They kill each other, which is clearly insane, and yet, here's the thing. When it's something that really matters, they fight. I mean, they're lame morons for fighting. But they do. They never... They never quit. And so I guess I will keep fighting, too."

Maybe a bit long, but I love it so!

Data: [asking Worf to take care of Spot] He will need to be fed once a day. He prefers feline supplement number 25.

Worf: I understand.

Data: And he will require water. And you must provide him with a sandbox - and you must talk to him. Tell him he is a pretty cat, and a good cat...

Worf: I will feed him.

Data: Perhaps that will be enough.

[Helen tells James her boyfriend is cheating]
James: Well, if it makes you feel any better... do you see that bloke over there?
[Points to his friend at the end of the bar]
James: Not only does he own a personalized matching set of crocodile-skin luggage, but his favorite TV program is Baywatch. So you see there's always someone sadder than you.
[Helen starts to cry]
James: Do you love him?
Helen: No, I could never love a Baywatch fan.
